Epinephrine Stimulation of Glucose Release from Perfused Trout Liver: Effects of Assay and Acclimation Temperature

چکیده
Since fish are poikilothermic, changes in temperature may perturb hormonal activation of cell function. To test this hypothesis, and determine the extent to which hormonal responses are thermally compensated, the effect of temperature on epinephrinestimulated glucose release in perfused trout liver was studied. Thermally acclimated (5 and 20 ̊C) rainbow trout (Oncorhynchus mykiss) were responsive to epinephrine (0.531026 mol l–1) at both 5 and 20 ̊C. Metoprolol (b2 antagonist) and propranolol (b1and2) decreased the response significantly (to 1.4% and 8.4% of stimulated values, respectively) while phentolamine (a1and2) was without effect, implying the response is b2-mediated. Both basal (86 and 19mmol g21 liver h21 in 5 and 20 ̊C trout, respectively) and epinephrine-stimulated (210 and 168 mmolg21 h21) rates of glucose release were higher (2.4-fold higher for epinephrine-stimulated and 8.8-fold for basal) in 5 ̊Cthan in 20 ̊C-acclimated fish, regardless of perfusion temperature. Although the dose–response curve for epinephrine was markedly temperature-dependent, coldand warm-acclimated fish were affected in different ways. Cold-acclimated fish (5 ̊C) were less responsive to epinephrine when perfused at 5 ̊C (ED50 6.831029 mol l–1) than when perfused at 20 ̊C (ED50 8.2310210 mol l–1); in contrast, warm-acclimated fish (20 ̊C) were less responsive to epinephrine when perfused at 20 ̊C (ED50 4.631027 mol l–1) than at 5 ̊C (ED50 6.631029 mol l–1). These results are interpreted as being indicative of adaptations to maintain the capacity for hepatic glucose mobilization at low temperature.

Thermal Acclimation Alters Both Adrenergic Sensitivity and Adrenoceptor Density in Cardiac Tissue of Rainbow Trout
We examined the effect of temperature acclimation on the sensitivity of the rainbow trout heart to adrenaline and on the density of b-adrenoceptors. The sensitivity of the heart was assessed using in situ working perfused heart and in vitro isometric ventricular strip preparations. When tested in situ and at acclimation temperature, hearts from fish acclimated to 8 ̊C were approximately 10-fold ...

The effects of glucagon and epinephrine on gluconeogenesis and glycogenolysis in liver and of catecholamines and adrenocorticotrophic hormone on lipolysis in adipose tissue were examined in normal and adrenalectomized rats. Glucagon or epinephrine stimulation of glucose synthesis from lactate was markedly reduced in isolated perfused livers from fasted, adrenal-deficient rats. Administration in...
متن کاملEffects of insulin-like growth factor I on basal and stimulated glucose fluxes in rat liver.
Effects of insulin-like growth factor I (IGF-I) and insulin on glucose and potassium fluxes were examined by measuring transhepatic glucose and potassium balance in isolated perfused rat livers. At 1 nM, both IGF-I and insulin decreased basal glucose release by approximately 64% (P < 0.05). Adrenaline (epinephrine)-stimulated glucose release (42.6 +/- 4.5 micromol/g of liver within 30 min) was ...
متن کاملCoronary flow in a perfused rainbow trout heart.
A preparation was developed to perfuse the coronary circulation in working hearts from rainbow trout (Salmo gairdneri Richardson). The preparation was used to examine pressure-flow relationships for the coronary circulation as the heart generated physiological and subphysiological work loads. Coronary vascular resistance increased exponentially as coronary flow rate decreased. Coronary resistan...
متن کاملGlucagon secretion from the perfused rat pancreas. Studies with glucose and catecholamines.
The isolated in situ perfused rat pancreas was used to study glucose and catecholamine control of glucagon secretion, and to investigate the possible role of endogenous cyclic AMP as a mediator of this secretory process. When perfusate glucose was acutely dropped from 100 to 25 mg/100 ml, glucagon was released in a biphasic pattern with an early spike and a later plateau-like response. 300 mg/1...
